Introduction
TFPIisaproteaseinhibitorwhichcontrolsthetissuefactor(TF)-dependentpathwayofbloodcoagulation.ThecoagulationprocessstartswiththecreationofafactorVIIa-TFcomplex,thatproteolyticallytriggersadditionalproteases(factorsIXandX)andeventuallyresultsinafibrinclot.TFPIinhibitstheactivatedfactorXandVIIa-TFproteasesinanautoregulatoryloop.TFPIisglycosylatedandpredominantlylocatedinthevascularendotheliumandplasmainbothfreeformsandcomplexedwithplasmalipoproteins.Anumberofalternativelysplicedtranscriptvariantsofthisgenehaveareknown,howeverthefull-lengthnatureofseveralofthesevariantswerenotyetestablished.

TFPIHumanRecombinantproducedinSf9Baculoviruscellsisasingle,glycosylatedpolypeptidechaincontaining285aminoacids(29-304a.a.)andhavingamolecularmassof33kDa(MolecularsizeonSDS-PAGEwillappearatapproximately40-57kDa).TFPIisexpressedwitha9aminoacidsHistagatC-Terminusandpurifiedbyproprietarychromatographictechniques.
