Introduction

VitaminDreceptors(VDRs)belongtotheNR1Ifamily,whichalsoincludespregnaneX(PXR)andconstitutiveandrostane(CAR)receptorswhichformheterodimerswithmembersoftheretinoidXreceptorfamily.VDRisexpressedintheintestine,thyroidandkidneyandhasanimperativeroleincalciumhomeostasis.VDRsinhibitexpressionof1alpha-hydroxylase(theproximalactivatorof1,25(OH)2D3andinduceexpressionofthe1,25(OH)2D3inactivatingenzymeCYP24.Additionally,VDRhasrecentlybeenrecognizedasanadditionalbileacidreceptoralongsideFXRandmayfunctiontoprotectgutagainstthetoxicandcarcinogeniceffectsoftheseendobiotics.HereditarymutationsintheVDRgeneleadstorickets,whichistypifiedbymuscleweakness,growthretardation,bonedeformityandsecondaryhyperparathyroidism.ThehumangeneencodingtheVDRislocalizedtochromosome12q12-q14.
