Introduction
FibroblastGrowthFactors(FGFs)compriseafamilyofatleasteighteenstructurallyrealtedproteinsthatareinvolvedinamultitudeofphysiologicalandpathologicalcellularprocesses,includingcellgrowth,differentation,angiogenesis,woundhealingandtumorgenesis.TheBIOLOGicalactivitiesoftheFGFsaremediatedbyafamilyiftypeItransmembranetyrosinekinaseswhichundergodimerizationandautophosphorylationafterligandbinding.FourdistinctgenesencodingcloselyrelatedFGFreceptors,FGFR-1to-4areknown.MultipleformsofFGFR-1to-3aregeneratedbyalternativesplicingofthemRNAs.AfrequentsplicingeventinvolvingFGFR-1and-2resultsinreceptorscontainingallthreeIgdomains,referredtoasthealphaisoform,oronlyIgIIandIgIII,referredtoasthe?isoform.OnlythealphaisoformhasbeenidentifiedforFGFR-3andFGFR-4.AdditionalsplicingeventsforFGFR-1to-3,involvingtheC-terminalhalfoftheIgIIIdomainencodedbytwomutuallyexclusivealternativeexons,generateFGFreceptorswithalternativeIgIIIdomains(IIIbandIIIc).AIIIaisoformwhichisasecretedFGFbindingproteincontainingonlytheN-terminalhalfoftheIgIIIdomainplussomeintronsequenceshasalsobeenreportedforFGFR-1.MutationsinFGFR-1to-3havebeenfoundinpatientswithbirthdefectsinvolvingcraniosynostosis.
