ConnectiveTissueGrowthFactorbelongstotheCCNfamilyofproteins.TheCCNfamilypresentlyconsistsofsixmembersinhumanalsoknownas:Cyr61(Cysteinrich61),CTGF(ConnectiveTissueGrowthFactor),Nov(NephroblastomaOverexpressedgene),WISP-1,2and3(Wnt-1InducedSecretedProteins).TheCCNgenesencodesecretedproteinsassociatedwiththeExtracellularMatrix(ECM)andcellmembrane.CCNproteinsarematricellularproteinswhichareinvolvedintheregulationofvariouscellularfunctionsincluding:proliferation,differentiation,survival,adhesionandmigration.Theyareexpressedinderivativesofthethreeembryonicsheetsandareimplicatedinthedevelopmentofkidney,nervoussystem,muscle,bonemarrow,cartilageandbone.Duringadulthood,theyareimplicatedinwoundhealing,bonefracturerepair,andpathologiessuchas:fibrosis,vascularailmentsandtumOrigenesis.FulllengthsecretedCCNproteinscanshowanantiproliferativeactivity,whereastruncatedisoformsarelikelytostimulateproliferationandbehaveasoncogenes.ThefulllengthproteinconsistsoffourmodulesModuleIsharespartialidentitywiththeN-terminalpartoftheInsulin-likeGrowthFactorBindingProteins(IGFBPs).ModuleIIincludesastretchof70aminoacidresidues–whichsharessequenceidentitywiththeVonWillebrandFactorTypeCrepeat(VWC).ModuleIIIcontainssequencessharingidentitywiththeThrombospondintype1repeat(TSP1)(WSXCSXXCG),whichisthoughttobeimplicatedinthebindingofsulfatedglycoconjugatesandtobeimportantforcelladhesion.ModuleIV,alsodesignatedCT,isencodedbyexon5.Itistheleastsconservedoneofthefourdomainsatthelevelofnucleotidesequence,butitappearstobecriticalforseveraloftheBIOLOGicalfunctionsattributedtotheCCNproteins.ModuleIVresemblestheCTdomainofseveralextracellularproteinincluding,VonWillebrand"sfactorandmucins.Sequencesimilaritiestoheparin-bindingmotifsarealsofoundwithinthisdomain.Proteolysisofthesecretedfull-lengthCCNproteinsthathasbeenreportedinthecaseofCCN2andCCN3mightresultintheproductionofCCN-derivedpeptideswithhighaffinityforligandsthatfull-lengthCNNproteinsbindonlypoorly.Amino-truncatedCCN2isoformswerebiologicallyactivewhereasnospecificbiologicalactivityhasbeenattributedtothetruncatedCCN3.Althoughthemolecularprocessesunderlyingtheproductionofthesesecretedisoformsispresentlyunknown,itisimportanttonotethatproteolysisoccuratthesameaminoacidresiduesinbothCCN2andCCN3.AnelevatedexpressionofCCN2hasalsobeendetectedbyNorthernblottinginhumaninvasivemammaryductalcarcinomas,dermatofibromas,pyogenicgranuloma,endothelialcellsofangiolipomasandangioleiomyomas,andinpancreatictumors.AstudyperformedwithchondrosarcomasrepresentativeofvarioushistologicalgradesestablishedthatCCN2expressionwascloselycorrelatedwithincreasinglevelsofmalignancy.InagreementwithCCN2playingaroleinbraintumorangiogenesis,immunocytochemistrystudiesindicatedthatbothglioblastomatumorcellsandproliferatingendothelialcellsstainedpositiveforCCN2.Inastrocytomas,CCN2expressionwasparticularlyelevatedinhighgradetumors,withamarkedeffectofCCN2oncellproliferation.DownregulationofCCN2expressioninthesecellswasassociatedwithagrowtharrestattheG1/Stransitionwhileover-expressionofCCN2inducedatwo-foldincreaseofthenumberofcellsintheG1phase.Geneprofilinganalysisallowedtoidentifyasetofabout50geneswhoseexpressionmightaccountfortheproliferativeactivityofCCN2inthesecells.CCN2wasseeninahigherproportionofmononuclearcellsofpatientswithacutelymphoblasticleukemia.
