Introduction
High-mobilitygroupbox1protein(HMGB1),previouslyknownasHMG-1oramphoterin,isamemberofthehighmobilitygroupboxfamilyofnon-histonechromosomalproteins.HumanHMGB1isexpressedasa30kDa,215aminoacid(aa)singlechainpolypeptidecontainingthreedomains:twoN-terminalglobular,70aapositivelychargedDNA-bindingdomains(HMGboxesAandB),andanegativelycharged30aaC-terminalregionthatcontainsonlyAspandGlu.4,5Residues27-43and178-184containaNLS.Posttranslationalmodificationsofthemoleculehavebeenreported,withacetylationoccurringonasmanyas17lysineresidues.HMGB1isexpressedathighlevelsinalmostallcells.ItwasoriginallydiscoveredasanuclearproteinthatcouldbendDNA.Suchbendingst
