ChikungunyaisaninfectioncausedbythechikungunyaviruswhichispassedtohumansbytwospeciesofmosquitoofthegenusAedes:A.albopictusandA.aegypti.Animalreservoirsofthevirusincludemonkeys,birds,cattle,androdents.Thefeaturesofthediseaseareasuddenonsetoffever2-4daysafterexposure.Thefevertypicallylasts2-7days,whiletheassociatedjointpainsusuallylastweeksormonthsbutsometimesyears.Themortalityrateisalittlelessthan1in1,000.ThediseasehasoccurredinoutbreaksinAsia,EuropeandtheAmericassince2004.CHIKVisasingle-strandedpositive-senseRNAgenome,11,800ntslongwhichencodes2openreADIngframes.Thenucleocapsidistightlyenvelopedbyahost-derivedlipidbilayer(envelope)supportingthevirus-encodedenvelopeproteins.80glycoproteinspikesareC-terminallyanchoredwithintheviralenvelope.ThestructuralpolyproteinistranslatedfromaviralsubgenomicmRNA,whileasthe5structuralproteins(capsid,E3,E2,6K,E1)aretranslatedasasinglepolyprotein,fromwhichcapsid(C)iscleavedofftoencapsidate.TheenvelopepolyproteinprecursorE3-E2-6K-E1istranslocatedtotheendoplasmaticreticulum.Polyproteinisprocessedbyhostsignalases,resultinginE3,E2&E1formingviralhetero-trimericspikes.TheviralspikesmajorlycontainsE2andE1facilitatecellreceptorrecognition,cellentrythrupH-dependentendocytosisandsupportviralbudding.
