TARCCDNAencodesa94aminoacidprecursorproteinwitha23aminoacidresiduesignalpeptidethatiscleavedofftogeneratethe71aminoacidresiduematuresecretedprotein.AlongwithCCchemokinefamilymembers,CCL-17hasapproximately24-29%aminoacidsequenceidentitywithRANTES,MIP-1a,MIP-1b,MCP-1,MCP-2,MCP-3andI-309.TARCisexpressedinthymus,andatalowerlevelinthelung,colon,andsmallintestine.TARCisinadditiontransientlyexpressedinstimulatedperipheralbloodmononuclearcells.RecombinantTARChasbeenshowntobechemotacticforTcelllinesbutnotmonocytesorneutrophils.CCL-17isaspecificfunctionalligandforCCR4,areceptorthatisselectivelyexpressedonTcells.CCL17isoneofafewCys-Cys(CC)cytokinegenesclusteredontheqarmofchromosome16.CCL17showschemotacticactivityforTlymphocytes,butnotmonocytesorgranulocytes.CCL17bindstochemokinereceptorsCCR4andCCR8.ThischemokineplaysimportantrolesinTcelldevelopmentinthymusaswellasintraffickingandactivationofmatureTcells.
