Introduction
NogginwhichisencodedbytheNOGgene,bindsandinactivatesmembersofthetransforminggrowthfactor-beta(TGF-beta)superfamilysignalingproteins,suchasbonemorphogeneticprotein-4(BMP4).BydiffusingthroughextracellularmatricesmoreefficientlythanmembersoftheTGF-betasuperfamily,nogginmayplayandimportantroleincreatingmorphogenicgrADIents.Nogginappearstohavepleiotropiceffect,bothearlyindevelopmentaswellasinlaterstages.NogginwasoriginallyisolatedfromXenopusbasedonitsABIlitytorestorenormaldorsal-ventralbodyaxisinembryosthathadbeenartificiallyventralizedbyUVtreatment.Theresultsofthemouseknockoutofnogginsuggestthatitisinvolvedinnumerousdevelopmentalprocesses,suchasneuraltubefusionandjointformation.ThereareseveraldominanthumanNOGmutationsinunrelatedfamilieswithproximalsymphalangism(SYM1)andmultiplesynostosessyndrome(SYNS1).AllNOGmutationsalteredevolutionarilyconservedaminoacidresidues.TheaminoacidsequenceofhumannogginishighlyhomologoustothatofXenopus,ratandmouse.

NogginproducedinSf9Baculoviruscellsisaglycosylatedhomodimercontaining205aminoacidsandhavingamolecularmassof47.9kDaundernon-reducingconditions.(MolecularsizeonSDS-PAGEwillappearatapproximately50-80kDa).Nogginispurifiedbyproprietarychromatographictechniques.
