Introduction
MCSFRwhichisalsofamiliarasCSF1R,ispartofthetype3subfamilyofreceptortyrosinekinases.MCSFRisexpressedmainlyoncellsofthemonocyteandmacrophagelineage,stemcells,andinthegrowingplacenta.MostoftheBIOLOGicaleffectsofthiscytokinearemediatedbyMCSFR.MCSFRcontainsanextracellularligand-bindingdomain,asinglemembrane-spanningsegment,andanintracellulartyrosinekinasedomain.OriginallyCSF1andthisreceptorwereimplicatedasvitalfornormaltrophoblasticimplantationaswellasmonocytedevelopment.TheroleofCSF1/CSF1RinnormalmammaryglanddevelopmentisactuallyveryinterestingsincethisconnectionhasalsobeendiscoveredinthebiologyofbreastcancerwiththeresultsofabnormalexpressionofCSF1anditsreceptor.Likewise,inAlzheimer"sdiseaseandafterbraininjuriesanincreasedlevelofCSF1Rwasfoundinthemicroglia,whichcausesthemicrogliatobecomemoreactive.

MCSFRproducedinSf9Baculoviruscellsisasingle,glycosylatedpolypeptidechaincontaining737aminoacids(20-517a.a.)andhavingamolecularmassof82.1kDa.(MolecularsizeonSDS-PAGEwillappearatapproximately70-100kDa).MCSFRisexpressedwitha239aminoacidhIgG-HistagatC-Terminusandpurifiedbyproprietarychromatographictechniques.
