IL-29isdistantlyrelatedtotypeIIFNsandtheIL-10family.ExpressionofIL-29isinducedbyviralinfectionwhichinteractswithaheterodimericclassIIcytokinereceptorthatconsistsofinterleukin10receptor,beta(IL10RB)andinterleukin28receptor,alpha.IL-29exhibitscommonfeatureswithtypeIIFNssuchasantiviralactivity,antiproliferativeactivityandinvivoantitumouractivity.IL-29actssimilarlytoIFNs,butislesseffectivegenerallyandhasactivityinamorelimitedrangeofcelllines.IFN-ambda1,IFN-lambda2andIFN-lambda3arecloselypositionedgenesonhumanchromosome19.IL-29inducesELR(-)CXCchemokinemRNAinhumanperipheralbloodmononuclearcells,inanIFN-gamma-independentmanner.IL-29isabletogeneratetolerogenicDCs,anactivitythatcouldthwartIFN-betafunctions.IL-29producedinresponsetoviralinfection,activatesbothmonocytesandmacrophagesproducingarestrictedpanelofcytokinesandthereforeisanimportantfactorinactivatinginnateimmuneresponsesatthesiteofviralinfection.IFN-Lambda1antiviralandantiproliferativeactivityrequiresIFN-Lambda2receptortyrosineresidues.
