Introduction
MembersoftheIFNregulatoryfactor(IRF)familyregulategeneexpressioncriticaltoimmuneresponse,hemopoiesis,andproliferation.IRF-3isamemberoftheIRFfamily,andisdistinctfromotherfamilymembers.Itstranscriptionalactivityisregulatedsolelybyposttranslationalmodifications.Itplaysacrucialroleinactivationofinnateimmunityandinflammationinresponsetoviralinfection.IRF-3mediatesIFN-stimulatedresponseelement(isre)promoteractivation.Functionsasamolecularswitchforantiviralactivity.DsrnageneratedduringthecourseofanviralinfectionleadstoIRF3phosphorylationonthec-terminalserine/threoninecluster.Thisinducesaconformationalchange,leADIngtoitsdimerization,nuclearlocalizationandassociationwithcrebbindingprotein(crebbp)toformdsrna-activatedfactor1(draf1),acomplexwhichactivatesthetranscriptionofgenesunderthecontrolofisre.Thecomplexbindstotheieandprdiiiregionsontheifn-alphaandifn-betapromotersrespectively.IRF-3doesnothaveanytranscriptionactivationdomains.
