GSTP1isapolymorphicgeneencodingactive,functionallydifferentGSTP1variantproteinsthatarebelievedtofunctioninxenobioticmetabolismandhaveapartinsusceptibilitytocancer,andotherdiseases.GSTP1isaglutathioneS-transferasebelongingtothepiclass.GSTfamilyenzymesplayasignificantroleindetoxificationbycatalyzingtheconjugationofmanyhydrophobicandelectrophiliccompoundswithreducedglutathione.Baseduponthebiochemical,immunologicandstructuralpropertiesofthesolubleGSTstheyaregroupedinto4mainclasses:alpha,mu,pi,andtheta.TheGSTP1enzymeactsbycatalyzingthereactionofglutathionewithanacceptormoleculetoformaSulfur-substitutedglutathione.Thereactionsemployingglutathionecontributethetransformationofabroadrangeofelectrophiles,includingreactiveproductsoflipid,protein,carcinogens,therapeuticdrugs,environmentaltoxins,andproductsofoxidativestress.TheGSTP1inactivationthroughCpGhypermethylationisfrequentinpituitaryadenomasandmaybeafactorinaggressivepituitarytumorbehavior.GSTP1ismaybeatranscriptionaltargetofthep53tumorsuppressorgene.Single-nucleotidepolymorphisminGSTP1islinkedtomodifiedproteinbinding,whichinfluenceGSTP1"scontributiontocarcinogenanddrugmetabolism,andpossIBLydiseasepathogenesisand/ordrugresponse.GST-pimighthavecentralrolesinproliferationofandrogen-independenthumanprostatecancercells.
