Introduction
Humannorovirusisclassifiedintotwogroups,group1&group2.Norwalkvirusisthespecieswhichbelongstogroup1andwasdiscoveredin1968atOhio.Norovirusisafamiliarviruswhichcauseshumangastroenteritiswiththefollowingsymptomsvomiting,diarrheaandsickness.CDCreportrevealedthatthereare19-21millionAmericansinfectedbyNororvirusannuallywith800deaths,1in15peoplewithinfection.Aroundtheworld,thisvirusaffectsabout267millionpeopleandcausesover200,000deathseachyear;theselossesaremostlyinlessdevelopedcountriesandintheveryyoung,elderlyandimmuno-suppressedpopulation,though,mostcasesareself-limitedwithafullrecoverywithinjustafewdays.Norovirusisextremelycontagiousandcanspreadfromhumantohumanthroughinfectedfood,waterorcontaminatedsurfaces.TheoutbreaksusuallyoccurfromNovember-April,whilethepeakisinJanuary.NorovirusisapositivesenseRNAviruswith7.5kbnucleotides,encodingamajorstructuralproteinVP1with50-55kDa.ThefulllengthofVP1capsidcomprisestheinternalN-terminal,Hinge,shell(S)andprotruding(P)domains.Pdomainfrom225to520formsP1-P2-P1structure.Moreover,Pdomainhasareceptorbindingregionwhichrecognizeshumanhisto-bloodgroupantigens(HBGAs).PdomainexpressedinbacteriacanspontaneouslyformaPdimeaswellasaPparticleaggregatedby12Pdimmers.PparticledisplaysanincreasedbindingactivitytoHBGAshigherthanvirus-likeparticle(VLP)formedbythefull-lengthcapsid.Fornorovirusvaccinedevelopment,weconsiderPdomainasagoodcandidate.
