Introduction

H3N2isasubtypeoftheinfluenzaAvirus.Itsnamederivesfromtheformsofthetwokindsofproteinsonthesurfaceofitscoat,hemagglutinin(H)andneuraminidase(N).H3N2exchangesgenesforinternalproteinswithotherinfluenzasubtypes.H3N2hastendedtodominateinprevalenceoverH1N1,H1N2,andinfluenzaB.H3N2straindescendedfromH2N2byantigenicshift,inwhichgenesfrommultiplesubtypesre-assortedtoformanewvirus.BoththeH2N2andH3N2strainscontainedgenesfromavianinfluenzaviruses.H3N2virusesareabletoinfectmammalsandbirds.Inpigs,humans,andbirds,thevirushasmutatedintomanystrains.Hemagglutinin(HA)bindstosialicacid-containingreceptorsonthecellsurface,generatingtheattachmentofthevirusparticletothecell.HAhasavitalpartinthedeterminationofhostrangerestrictionandvirulenceandisinchargeofthediffusionofthevirusintothecellcytoplasmbyfacilitatingthefusionofthemembraneoftheendocytos

CanineH3N2producedinSf9Baculoviruscellsisasingle,glycosylatedpolypeptidechaincontaining336aminoacids(18-344a.a.)andhavingamolecularmassof36.9kDa.(MolecularsizeonSDS-PAGEwillappearatapproximately40-57kDa).

H3N2isexpressedwitha6aminoacidHistagatC-Terminusandpurifiedbyproprietarychromatographictechniques.
