Introduction
AntioxidantenzymeGlutathioneS-Transferase(GST)isthoughttodotheprimarycellulardefensemechanismagainstreactiveoxygenspecies.GSTreduceslipidhydroperoxidesthroughitsSe-independentglutathioneperoxidaseactivity.Theenzymealsodetoxifieslipidperoxidationendproductssuchas4-hydroxynonenal(4-HNE).ThesolubleGSTisa26kDaproteinwhichoccursasadimerinallaerobicorganisms.Eachmonomerhastwodomains,onethatbindsGSHandisan/-structuresimilartothioredoxinandtheother,allhelical,thatbindsthehydrophobicsubstrate.TheGST-fusionproteinexpressionsystemisawidelyusedrecombinantproteinexpressionsystemthatallowsapeptideoraregulatoryproteindomaintobeexpressedasafusiontotheC-terminusofSchistosomajaponicumGST.FusionproteinsalsopossessGST-enzymaticactivityandcanundergodimerizationsimilartoinvivo.ThefusionproteincanbepurifiedviaGST-affinitycolumnchromatography.Inmostcases,thedesiredpeptidesordomainsareremovedfromGSTbyapplyingaspecificproteasethatrecognizesandcleavesthelinkerbetweentheproteindomainandGST.Thetechniquehasbeenwidelyusedtogeneratedifferentkindsofproteinsforcrystallization,molecularimmunologystudies,theproductionofvaccinesandstudiesinvolvingprotein-proteinandprotein-DNAinteractions.
