GNMTisanenzymethatcatalyzestheconversionofS-adenosyl-L-methioninewithglycinetoS-adenosyl-L-homocysteineandsarcosine.GNMTislocatedinthecytoplasmandactsasahomotetramer.DefectsintheGNMTgenecausesofGNMTdeficiency(hypermethioninemia).GNMTaffectsDNAmethylationbyregulatingtheratioofS-adenosylmethioninetoS-adenosylhomocystineandisinvolvedinthedetoxificationpathwayinlivercells.GNMTexpressionisdiminishedinhumanhepatocellularcarcinoma(HCC).GNMTcatalyzesthemethylationofglycinebyusings-adenosylmethionine(adomet)toformn-methylglycine(sarcosine)withtheconcomitantproductionofs-adenosylhomocysteine(adohcy).GNMTplaysanessentialroleintheregulationoftissueconcentrationofadometandofmetabolismofmethionine.
