Theepidermalgrowthfactorreceptor(EGFR)subfamilyofreceptortyrosinekinasescomprisesfourmembers:EGFR(alsoknownasHER1,ErbB1orErbB),ErbB2(Neu,HER-2),ErbB3(HER-3),andErbB4(HER-4).AllfamilymembersaretypeItransmembraneglycoproteinthathasanextracellulardomainwhichcontainstwocysteine-richdomainsseparatedbyaspacerregionthatisinvolvedinligand-binding,andacytoplasmicdomainwhichhasamembrane-proximaltyrosinekinasedomainandaC-terminaltailwithmultipletyrosineautophosphorylationsites.ThehumanEGFRgeneencodesa1210aminoacid(aa)residueprecursorwitha24aaputativesignalpeptide,a621aaextracellulardomain,a23aatransmembranedomain,anda542aacytoplasmicdomain.EGFRhasbeenshowntobindasubsetoftheEGFfamilyligands,includingEGF,amphiregulin,TGF-a,betacellulin,epiregulin,HBEGFandneuregulin-2intheabsenceofaco-receptor.LigandbindinginducesEGFRhomodimerizationaswellasheterdimerizationwithErbB2,resultinginkinaseactivation,tyrosinephosphorylationandcellsignaling.EGFRcanalsoberecruitedtoformheterodimerswiththeligand-activatedErbB3orErbB4.EGFRsignalinghasbeenshowntoregulatemultipleBIOLOGicalfunctionsincludingcellproliferation,differentiation,motilityandapoptosis.Inaddition,EGFRsignalinghasalsobeenshowntoplayaroleincarcinogenesis.
