Leptinisaproteinhormone.ItismainlyproducedinADIposecellsthatregulateenergyhomeostasisbyrestraininghunger.Leptintiestonuclearreceptorsinthehypothalamus(arcuatenucleus).SimilartoinsulinresistanceintypeIIdiabetes,inobesitythereisadecreaseinthesensitivitytowardsleptin,endinginafailuretoidentifysatiety,eveninhighlevelsofenergystoresorleptinitself.Full-lengthCDNAencoding2leptinsequences(tLepAandtLepB)and1leptinreceptorsequence(tLepR)existsintilapia(Oreochromisniloticus).Thefull-lengthcDNAoftLepRis3423bp,encodingaproteinof1140aminoacidwhichcontainedallfunctionallyimportantdomainsconservedamongvertebrateleptinreceptors.ThecDNAsoftLepAandtLepBare486bpand459bpinlength,encodingproteinsof161aaand152aa,respectively.Thethree-dimensionalstructuresoftLepAandtLepBdemonstratesstrongconservationoftertiarystructurewiththatofhumanleptincomprisedof4helixes.
